Generated in rabbit and affinity-purified against a synthetic phosphopeptide centered on the serine 1083 site of human SMC3, this polyclonal primary antibody from Antibodies.com is supplied at 1 mg/ml in phosphate buffered saline containing 15 mM sodium azide, pH approximately 7.2. SMC3 (Structural Maintenance of Chromosomes 3) is the kleisin-associated ATPase subunit of the cohesin ring that physically entraps sister chromatids from S phase until anaphase onset, and modification of serine 1083 contributes to the regulatory program that releases cohesin from chromatin during mitotic exit and following DNA double-strand breaks.

Validated for Western blot on whole-cell lysates prepared from human, mouse, and rat samples, the unconjugated reagent is shipped as a 100 µl vial and is intended for research use only. Investigators studying chromosome segregation, sister chromatid cohesion, the DNA damage response, ATM/ATR signaling, and cell cycle checkpoints can use it to discriminate the phosphorylated form of SMC3 from the total protein pool in mitotically arrested cells, in cells released from a double-thymidine block, or in cultures exposed to ionizing radiation and other genotoxic agents that activate the DNA damage checkpoint.
