Smad5 is a receptor-regulated SMAD that transduces bone morphogenetic protein (BMP) signals downstream of BMP type I receptors, forming transcriptional complexes with SMAD1, SMAD9, and SMAD4 to regulate genes controlling osteogenic differentiation and hepcidin-mediated iron homeostasis. Antibodies.com distributes this mouse monoclonal antibody, raised against the C-terminal region of human Smad5, as a 100 µL vial of purified immunoglobulin in 0.1 M Tris-Glycine buffer (pH 7.4) containing 150 mM NaCl, 0.2% sodium azide, and 50% glycerol.

The unconjugated antibody is validated for Western blot, immunocytochemistry (ICC), and flow cytometry (FC), with reactivity confirmed on human and mouse samples. Researchers use this reagent to interrogate BMP-driven transcriptional outputs in cell culture, map SMAD phosphorylation dynamics in tissue lysates, and characterize SMAD1/5/9 complex assembly in chromatin immunoprecipitation experiments. As a C-terminal epitope reagent, it recognizes the C-terminal portion of Smad5 and is suited to detecting the full-length protein in fixed cells and whole-cell extracts.

For short-term use, store refrigerated; for longer storage, freeze aliquots at -20 °C and limit freeze-thaw cycles to preserve binding activity. The monoclonal format supports lot-to-lot reproducibility across experiments, and the 0.2% sodium azide preservative inhibits microbial contamination during repeated handling at the bench.
