This recombinant rabbit monoclonal antibody specifically detects Smad3 phosphorylated at serine residues 423 and 425, the activating modification introduced by the TGF-β type I receptor (ALK5) within the canonical SMAD signaling cascade. Researchers apply it to track receptor-driven SMAD3 activation in pathways governing cell proliferation, extracellular matrix deposition, and fibrotic responses across disease models.

Validated for Western blot, immunocytochemistry/immunofluorescence, and immunohistochemistry, the unconjugated reagent from Antibodies.com reacts with Human and Mouse samples, supporting translational studies in oncology, renal and pulmonary fibrosis, and developmental biology. The antibody is supplied as a 100 µl vial, formulated in 1×TBS (pH 7.4) with 1% BSA, 40% glycerol, and 0.05% sodium azide as a preservative. Shipped refrigerated; store at -20 °C and protect from light, avoiding repeated freeze-thaw cycles to preserve phospho-specific binding activity.

This reagent distinguishes the activated, transcription-competent form of SMAD3 from the total protein pool, enabling time-course analyses of TGF-β signal transduction in paraffin-embedded tissue, cultured cell lines, and primary cell lysates.
