Smad3 is a receptor-regulated intracellular transducer of the TGF-β signaling cascade, forming complexes with Smad4 that translocate into the nucleus to modulate transcription of target genes involved in fibrosis, epithelial-mesenchymal transition, and growth suppression. The M200 epitope lies in the C-terminal region of the protein, allowing detection of full-length Smad3 as well as C-terminally phosphorylated forms that arise after TGF-β receptor activation.

This unconjugated rabbit polyclonal reagent is supplied at 1 mg/ml in phosphate buffered saline (PBS) containing 15 mM sodium azide at approximately pH 7.2, and is validated for western blot applications across human, mouse, and rat samples. Researchers investigating TGF-β-dependent pathways in cancer biology, fibrotic disease models, and stem cell differentiation commonly use anti-Smad3 antibodies to monitor pathway activation status, since phosphorylation of Smad3 at the C-terminal SSVS motif is a direct readout of ligand-driven signaling.

The 100 µl pack size supports multiple gel runs and time-course experiments, and the polyclonal format increases the likelihood of recognizing Smad3 even when samples contain closely related family members such as Smad1, Smad2, or Smad5. From Antibodies.com, this reagent is intended for research use only and should be handled with appropriate sodium azide precautions.
