Anti-SMAD2 + SMAD3 Antibody

Raised in rabbit and affinity purified, this unconjugated IgG polyclonal reagent from Antibodies.com recognizes both SMAD2 and SMAD3, the receptor-regulated R-SMADs that transduce TGF-β signals from the membrane into the nucleus. The immunogen is a synthetic phosphorylated peptide corresponding to the C-terminal Ser423 and Ser425 motif of human SMAD3 (and the equivalent Ser465/Ser467 region of SMAD2), the canonical type I receptor phosphorylation site used as a direct read-out of pathway activation.

Validated for western blot, immunohistochemistry, and immunofluorescence on human, mouse, and rat samples, the dual-specificity format simplifies side-by-side comparison of SMAD2 versus SMAD3 engagement in fibrosis models, tumor cell lines, and developmental tissues. On western blot the reagent resolves the two endogenous bands at 52 kDa (SMAD2) and 60 kDa (SMAD3), with a recommended working dilution of 1:500 to 1:2,000 depending on antigen abundance and detection chemistry.

Each vial contains 100 µl of liquid antiserum formulated in PBS (pH 7.3) with 50% glycerol and 0.01% Thiomersal as preservative. Aliquot the stock on first use, store refrigerated at 4°C protected from light, and limit freeze-thaw cycles of the working solution; for long-term retention freeze at -20°C.
