Description

This unconjugated rabbit polyclonal reagent recognizes the activated, doubly phosphorylated C-terminal motifs of the receptor-regulated SMAD transcriptional effectors: SMAD2 (phospho-S465 + S467) and SMAD3 (phospho-S423 + S425). Phosphorylation at these paired serines is carried out directly by the type I TGF-β receptor (TβRI/ALK5) and related activin and nodal receptors, so the antibody serves as a read-out of canonical TGF-β family pathway activation rather than total SMAD protein levels.

Supplied from Antibodies.com as 200µl of IgG in a standard vial, the antibody is validated for western blot application and has been confirmed reactive with human and mouse samples. Researchers apply this reagent to lysates from cell culture systems stimulated with TGF-β1, TGF-β2, or TGF-β3 to follow receptor-mediated SMAD activation kinetics, to monitor epithelial-mesenchymal transition programs, and to study fibrosis, cancer, and developmental biology questions. Because the epitope is phosphorylation-dependent, phosphatase inhibitors should be included in lysis buffers and blot membranes handled without harsh stripping reagents that remove the phospho-signal.

Aliquot upon arrival and store at -20 °C; avoid repeated freeze-thaw cycles that degrade antibody activity over time. For research use only.
