Phosphorylation of Smad5 at serine residues 463 and 465 marks its activation downstream of bone morphogenetic protein (BMP) type I receptors, including ALK2, ALK3, and ALK6. This rabbit monoclonal antibody, supplied by Antibodies.com, selectively recognizes the dually phosphorylated form of Smad5 (phospho-S463/465) while showing no appreciable reactivity against the non-phosphorylated protein. It is validated for Western blot, immunocytochemistry, and immunohistochemistry applications across human, mouse, and rat samples, making it a versatile reagent for studies in BMP signaling, osteoblast differentiation, vascular biology, and hepcidin-mediated iron regulation.

The reagent is supplied as 100 µl of recombinant rabbit monoclonal antibody in 1× TBS (pH 7.4) supplemented with 1% BSA, 40% glycerol, and 0.05% sodium azide as preservative. For routine use, store refrigerated at 4 °C; for extended storage, aliquot and freeze at -20 °C, avoiding repeated freeze-thaw cycles that can degrade antibody activity. This unconjugated format allows flexible pairing with HRP-, fluorophore-, or biotin-conjugated secondary reagents for chemiluminescent, fluorescent, or chromogenic detection workflows. Investigators studying BMP-driven processes such as bone formation, chondrogenesis, iron homeostasis, or hereditary hemorrhagic telangiectasia can apply this reagent to monitor endogenous Smad5 activation in cell lysates, cultured cells, and fixed tissue sections.
