The SHMT1 target is serine hydroxymethyltransferase 1, a cytosolic enzyme central to one-carbon folate metabolism. It catalyzes the reversible conversion of serine to glycine while generating 5,10-methylenetetrahydrofolate, feeding carbon units into nucleotide biosynthesis and methylation pathways. Aberrant SHMT1 expression has been linked to proliferation phenotypes in oncology and to disorders of folate-dependent neural tube development.
