Description

This rabbit polyclonal IgG antibody from Antibodies.com specifically detects SHC1 phosphorylated at tyrosine 427 (pY427), a regulatory phosphorylation site on the SHC1 adaptor protein. Phosphorylation at this tyrosine is triggered by activated receptor tyrosine kinases and other upstream signals, modulating SHC1's interactions with GRB2 and downstream propagation through the Ras-MAPK cascade during growth factor, integrin, and oxidative stress responses.

The unconjugated reagent is supplied as a 50 µl volume of polyclonal IgG and has been validated for western blot, immunohistochemistry, and immunofluorescence workflows. Reactivity has been confirmed in human, mouse, and rat samples, supporting comparative signaling studies across standard laboratory model systems.

Because recognition depends on a phospho-tyrosine modification rather than linear sequence alone, lysates should be prepared with phosphatase inhibitors to preserve phosphorylation. Working dilutions should be titrated independently for each application format, including standard western blot membranes, fixed tissue sections for IHC, and fixed cell preparations for IF imaging.
