Supplied by Antibodies.com as catalog number A39347, this Rabbit polyclonal reagent is raised against Shc1 (Ab-349) and is offered in a 50 µl format at a concentration of 1.0 mg/mL. The host species is Rabbit, the clonality is Polyclonal, and the antibody is supplied Unconjugated, making it suitable for downstream labelling workflows. It has been validated for two principal applications, namely Western blotting (WB) and immunocytochemistry/immunofluorescence (ICC/IF), and the recommended working dilutions are typically 1:500–1:1000 for WB and 1:100–1:200 for IF on methanol-fixed cells, as demonstrated on lysed HepG2 extracts and HeLa cells. The immunogen corresponds to a synthetic peptide spanning amino acids 347–351 (H-Q-Y-Y-N) of human Shc1, and the antibody specifically recognises the endogenous level of total Shc1 protein, with predicted bands at approximately 46, 52 and 66 kDa corresponding to the multiple Shc1 isoforms. Reactivity has been confirmed in Human, Mouse and Rat samples, covering the species most commonly used in signalling research. The buffer formulation is phosphate-buffered saline (without Mg²⁺ and Ca²⁺), pH 7.4, containing 150 mM NaCl, 0.02% sodium azide and 50% glycerol as a cryoprotectant. For storage and shipping, the vial is dispatched at 4 °C and, upon receipt, should be aliquoted and stored at –20 °C, with repeated freeze/thaw cycles avoided to preserve activity; under these conditions the reagent remains stable for long-term use. The target, SHC1 (Gene ID 6464, UniProt P29353), is also known as SH2 domain protein C1, SHC-transforming protein 1/3, SHCA and Src homology 2 domain-containing-transforming protein C1, an adaptor that couples activated receptor tyrosine kinases to Ras signalling. This product is intended for research use only and is not for diagnostic or therapeutic applications.
