Anti-SH3KBP1 Antibody

Raised in rabbit against the SH3KBP1 adaptor (also called CIN85), this unconjugated polyclonal IgG reagent from Antibodies.com is supplied as 100 µl of liquid antiserum in a single-use vial and is validated for Western blot detection of endogenous SH3KBP1 in human and mouse samples. The target is a multifunctional scaffold of roughly 85 kDa built from three N-terminal SH3 domains that recognize proline-rich ligands—including the Cbl family of E3 ubiquitin ligases and the endophilins—and a C-terminal coiled-coil that drives homotrimerization. Researchers use this reagent to probe CIN85-dependent trafficking of receptor tyrosine kinases such as EGFR and c-Met through the Cbl–CIN85–endophilin clathrin-coated-pit complex, to monitor Src-mediated CIN85 phosphorylation that gates receptor ubiquitination and sorting into multivesicular bodies, and to investigate how CIN85 oligomerization with SLP-65 sets the threshold for B-cell receptor activation and links BCR signals to the canonical NF-κB pathway via IKK-β. The antibody has also been used in immunoblot studies of patient fibroblasts and B-cell lines carrying X-linked germline deletions in CIN85 that cause antibody deficiency (immunodeficiency 61).
