SDHB is anchored in the inner mitochondrial membrane, where it channels electrons from succinate oxidation through its iron-sulfur clusters to ubiquinone, coupling the TCA cycle to the electron transport chain. Loss of SDHB expression — typically visualized as an absent band on western blot of patient tumor lysates — is a hallmark diagnostic feature of SDH-deficient paraganglioma, pheochromocytoma, and an established molecular subtype of renal cell carcinoma. The reagent is also widely used in basic mitochondrial biology and metabolic research to assess Complex II assembly and steady-state abundance.
