RPS6KA3 encodes the RSK2 (p90 ribosomal S6 kinase 2) protein, a serine/threonine kinase that operates downstream of the MAPK/ERK signaling pathway. This polyclonal antibody from Antibodies.com is raised in rabbit and is validated for Western blot (WB) and immunohistochemistry (IHC) applications. It is supplied as a 100 µl liquid IgG fraction, unconjugated, and has been confirmed to react with human and mouse samples.

RSK2 contains two distinct kinase domains and is activated by ERK1/2-mediated phosphorylation in response to growth factors, mitogens, and cellular stress. Once activated, the kinase phosphorylates transcription factors, components of the translational machinery, and histone H3, coupling extracellular signals to changes in gene expression and protein synthesis. Loss-of-function mutations in RPS6KA3 cause Coffin-Lowry syndrome, an X-linked intellectual disability disorder with skeletal and growth abnormalities, making this reagent a useful tool for studies of neuronal signaling, developmental biology, and kinase pathway research.

The unconjugated IgG format allows flexible pairing with horseradish peroxidase, alkaline phosphatase, or fluorescent secondary reagents depending on the chosen detection method. For Western blot, working dilutions between 1:500 and 1:2000 are commonly used, while IHC on fixed tissue sections is often performed at 1:100 to 1:500. Refrigerate the vial for short-term use; for longer storage, aliquot and freeze at -20°C, avoiding repeated freeze-thaw cycles.
