This phospho-specific polyclonal antibody from Antibodies.com selectively recognises ribosomal protein S6 (RPS6) when phosphorylated at serine 240, a modification catalysed by p70 S6 kinase (S6K1) downstream of mTORC1. The reagent is raised in rabbit and supplied as 100 µl of affinity-purified IgG at 1 mg/ml in phosphate-buffered saline containing 0.05% sodium azide at approximately pH 7.2, with no additional carrier protein.

Phosphorylation at S240 (typically co-occurring with the neighbouring S244 site) is one of the most widely used functional readouts of mTOR pathway activation. Researchers rely on this modification to monitor signalling involved in cap-dependent translation, ribosome biogenesis, cell growth and proliferation, nutrient sensing, and the regulation of autophagy. The antibody cross-reacts with human, mouse, and rat RPS6, and has been validated for western blot detection of the phospho-band migrating near 32 kDa, as well as for immunohistochemistry and immunofluorescence staining of cultured cells and tissue sections.

For best results, the working stock should be kept refrigerated and protected from light; long-term storage of aliquots in a frozen state is recommended to minimise activity loss from repeated freeze-thaw cycles. The sodium azide in the buffer makes the reagent compatible with standard immunostaining workflows without further addition of preservative.
