This unconjugated rabbit polyclonal antibody from Antibodies.com targets RNGTT (RNA guanylyltransferase and 5'-phosphatase), the bifunctional enzyme that builds the protective 5' cap on nascent pre-mRNA. Supplied as 100 µl of polyclonal antiserum raised in rabbit, the reagent is validated for Western blot and immunofluorescence workflows and detects endogenous capping enzyme in human, mouse, and rat samples.

RNGTT operates through two coordinated domains: its RNA 5'-triphosphatase activity removes the γ-phosphate from the 5' end of pre-mRNA, and its guanylyltransferase domain then transfers GMP from GTP to form the cap via a covalent enzyme-GMP intermediate. The cap structure shields transcripts from 5' exonucleolytic decay, recruits translation initiation factors such as eIF4E, and marks RNAs as host-derived to limit innate immune activation. Monitoring RNGTT therefore supports studies of transcription cycle dynamics, cap-dependent translation control, RNA processing factor localization, and innate immune sensing of uncapped transcripts.

Recommended uses include Western blot analysis of whole-cell lysates to compare expression across species or treatment conditions, and immunofluorescence on fixed cells and tissue sections to map the subcellular distribution of the capping machinery. Handle the unconjugated vial in frozen single-use aliquots to limit freeze-thaw cycles and protect from light, in line with standard polyclonal antibody storage practice.
