The RNF20 protein is the catalytic subunit of a conserved E3 ubiquitin ligase complex that monoubiquitinates histone H2B on lysine 120, a chromatin mark tightly coupled to RNA polymerase II elongation and to the cellular response to DNA damage. Working with its heterodimeric partner RNF40, RNF20 deposits H2BK120ub across actively transcribed loci and is required for proper expression of many inducible and developmental genes, as well as for efficient homologous recombination repair. Loss or knockdown of RNF20 therefore produces broad transcriptional changes and sensitises cells to genotoxic stress, making the protein a target of interest in chromatin biology, transcription regulation, and cancer signalling studies.
This recombinant monoclonal reagent from Antibodies.com is raised in rabbit and affinity-purified against human RNF20, with confirmed reactivity against both human and mouse samples. It is supplied unconjugated in 100 µL of TBS buffer (pH 7.4) containing 1% BSA, 40% glycerol, and 0.05% sodium azide as preservative, a format suited to standard immunodetection workflows. Validated applications include Western blotting of whole-cell and nuclear extracts, immunocytochemistry on cultured cells, and flow cytometry, allowing H2B ubiquitination pathway readouts, subcellular localisation studies, and quantitative expression analysis within a single reagent. Store refrigerated at 2-8 degrees C and avoid repeated freeze-thaw cycles to preserve activity.
The antibody is intended for research use only and performs well on denaturing gels, where a band near the predicted ~115 kDa molecular weight of human RNF20 is typically observed. Recommended starting dilutions vary by application, and parallel probing with anti-H2BK120ub or anti-RNF40 antibodies is a common strategy for investigating the RNF20/40-dependent ubiquitination axis in chromatin, transcription, and DNA damage response experiments.
