This unconjugated rabbit polyclonal IgG reagent from Antibodies.com recognizes Regulator of G-protein Signaling 9 (RGS9), a GTPase-activating protein that accelerates GTP hydrolysis on heterotrimeric G-protein alpha subunits and terminates G-protein signaling cascades.

RGS9 expresses two major alternatively spliced isoforms with very different distributions. RGS9-1 is restricted to retinal photoreceptors, where it deactivates transducin during the recovery phase of phototransduction. RGS9-2 is concentrated in the striatum, including the caudate-putamen and nucleus accumbens, where it modulates dopamine D2 receptor and mu-opioid receptor signaling. Loss-of-function mutations in RGS9 have been linked to bradyopsia, a condition characterized by slowed rod and cone recovery in humans, and RGS9-2 knockout mice display enhanced behavioral responses to dopaminergic agents and morphine.

Validated for Western blot on mouse samples, this reagent is used to probe endogenous RGS9 in tissue lysates such as striatum, retina, and other neural preparations. Standard workflows include homogenization of fresh or flash-frozen tissue in RIPA buffer, separation by SDS-PAGE, transfer to PVDF or nitrocellulose membranes, and probing at a dilution series to optimize signal-to-noise. The antibody supports comparative studies of isoform expression between wild-type and RGS9-deficient animals and is suited to investigations of G-protein deactivation kinetics in the visual and dopaminergic systems.

Supplied unconjugated in a 200 µl volume, the antibody should be stored at -20 °C in working aliquots to limit freeze-thaw cycles; for short-term use the vial may be kept refrigerated at 4 °C. Intended for research use only.
