The Anti-Rb (phospho Ser795) Antibody [28B5] from Antibodies.com is a mouse monoclonal antibody that recognizes the retinoblastoma tumor suppressor protein only when phosphorylated at serine 795. This phosphorylation site is a key regulatory mark introduced primarily by Cyclin D-CDK4/6 complexes during the G1 phase of the cell cycle, where it triggers release of E2F transcription factors and drives progression into S phase. The reagent is supplied as a liquid IgG2a preparation at 100 µg and is validated for Western blot and ELISA applications on human samples. Because the antibody is restricted to the phosphorylated epitope, it is widely used to monitor CDK4/6 pathway activity, cell cycle entry, and the pharmacological response to CDK4/6 inhibitors in cancer research. For Western blot, lysates are typically prepared with phosphatase inhibitors to preserve the phospho-Ser795 mark, separated by SDS-PAGE, and the antibody is generally used at a working concentration in the low microgram per milliliter range with a horseradish peroxidase-conjugated secondary antibody. Store the vial at -20°C in small aliquots and avoid repeated freeze-thaw cycles to maintain reagent performance. For research use only.
