Anti-RAMP2 Antibody

The antibody targets RAMP2 (Receptor Activity Modifying Protein 2), a single-pass transmembrane accessory protein that traffics with the calcitonin receptor-like receptor (CLR/CALCRL) to generate the functional adrenomedullin receptor (AM1) at the plasma membrane. RAMP2 is essential for cardiovascular and lymphatic vascular development, and global knockout mice die in utero from vascular permeability defects and edema.

This Rabbit polyclonal reagent from Antibodies.com is supplied as 100 µl of a 1 mg/ml stock in PBS with 0.1% Sodium Azide and 50% glycerol, a cryoprotective buffer that supports frozen storage at -20°C with minimal freeze-thaw damage. The unconjugated IgG has been validated for Western blot (WB) detection of human and mouse RAMP2, producing a band near the expected ~20 kDa apparent molecular weight. It is appropriate for immunoblotting of cell lysates from cultured endothelial lines such as HUVEC and HMVEC, and from mouse tissues including lung, heart, and lymph nodes.

For clean blots, load 20-40 µg of total protein per lane on a 12-15% SDS-PAGE gel, transfer to PVDF or nitrocellulose, and dilute to 0.5-2 µg/ml (1:500-1:2000 from the 1 mg/ml stock) in 5% non-fat milk/TBST for 1 hour at room temperature or overnight at 4°C. The 50% glycerol formulation allows working dilutions to be re-frozen and reused. For long-term storage, aliquot the reagent to limit repeated freeze-thaw cycles. The azide-containing buffer is incompatible with live-cell and in vivo workflows; remove the azide by dialysis or buffer exchange before any conjugation or animal work.
