This unconjugated rabbit polyclonal IgG antibody from Antibodies.com specifically recognizes RAF1 only when phosphorylated at serine 259, a key inhibitory regulatory site within the MAPK/ERK signaling cascade. Phosphorylation at S259 keeps the kinase sequestered in an inactive conformation through 14-3-3 protein binding and is reversed by protein phosphatase 2A once upstream RAS signaling is engaged, making phospho-state-selective detection at this residue a useful readout of pathway status.

Validated for western blot detection of endogenous human RAF1, the reagent is suited to lysates prepared from cultured mammalian cell lines. It performs reliably on whole-cell extracts generated with standard RIPA buffer, where the phospho-specific epitope lets researchers distinguish the inactive, AKT-phosphorylated pool of RAF1 from the activated kinase population that has been dephosphorylated at S259.

Supplied as 200 µl of liquid antiserum, the antibody pairs well with anti-pan-RAF1 or anti-phospho-MEK1/2 reagents for parallel pathway studies in oncology and signal transduction research. Human reactivity is reported by the manufacturer, and storing the reagent in small single-use aliquots helps preserve phospho-specific binding activity across multiple freeze-thaw cycles.
