This polyclonal antibody from Antibodies.com specifically recognizes human RAF1 phosphorylated at serine 259, a regulatory site within the MAPK/ERK signaling cascade. Phosphorylation at S259 by AKT/PKB holds RAF1 in an inactive cytosolic conformation, so the phospho-S259 form is a common readout for studying pathway activity upstream of MEK and ERK, including experiments where RAF1 activation is inferred from loss of the S259 mark on a parallel blot.

Raised in rabbit, the IgG reagent is supplied unconjugated in 100 µl volumes and validated for western blot on human samples. The polyclonal response targets the phosphorylated S259 epitope alongside neighbouring determinants, helping detection of the modified form even when local denaturation during SDS-PAGE reduces antibody binding to a strictly conformational site.

Recommended use: probe cell lysates or tissue extracts to separate the inactive, AKT-phosphorylated pool of RAF1 from total RAF1, supporting studies of growth-factor, oncogenic and stress-dependent regulation of the pathway. IgG, unconjugated, 100 µl format.
