Raf-1, also known as C-Raf or RAF1, is a serine/threonine kinase that operates within the Ras-Raf-MEK-ERK signaling cascade, where membrane-bound Ras recruits Raf-1 for activation, leading to downstream phosphorylation of MEK1/2 and ERK1/2 and ultimately to changes in cell proliferation, differentiation, and survival. This Antibodies.com reagent is a rabbit polyclonal antibody raised against the Q39 epitope region of human Raf-1 and is supplied at 1 mg/ml in phosphate buffered saline (PBS) containing 0.05% sodium azide at approximately pH 7.2.

Validated for western blot and immunohistochemistry applications, the unconjugated antibody recognizes Raf-1 in human, mouse, and rat samples, enabling cross-species comparison in standard research workflows. For western blot, 20-30 µg of total protein can be separated by SDS-PAGE, transferred to nitrocellulose or PVDF, and probed at working dilutions between 1:500 and 1:1000. For immunohistochemistry on formalin-fixed paraffin-embedded tissue, heat-induced epitope retrieval in 10 mM citrate buffer at pH 6.0 for 10-20 minutes generally restores antigenicity.

The polyclonal format provides broader epitope coverage than a monoclonal reagent and is therefore appropriate for detecting full-length Raf-1 alongside common post-translationally modified species. Each vial contains 50 µl of stock; aliquoting is recommended to limit repeated freeze-thaw cycles if the reagent will be used over an extended period. Brief centrifugation before opening will recover any material that may have collected at the cap or tube wall.
