Raf-1 (also known as C-Raf) is a serine/threonine kinase that initiates the canonical RAF/MEK/ERK signaling cascade downstream of activated Ras, and the polyclonal reagent distributed by Antibodies.com was raised in rabbit against an immunizing peptide designated Q39. Raf-1 contains an N-terminal regulatory region with the RAS-binding domain and cysteine-rich domain responsible for membrane recruitment, and a C-terminal kinase domain that phosphorylates MEK; the Q39 peptide directs the antibody to a defined portion of this larger protein. Intended for research use only.
Supplied as a 100 µl vial at 1 mg/ml in phosphate buffered saline (PBS) with 0.05% sodium azide at approximately pH 7.2, the unconjugated antibody is validated for Western blot (WB) and immunohistochemistry (IHC) workflows. It cross-reacts with human, mouse, and rat Raf-1, allowing consistent detection across cultured human cell lines, rodent model systems, and archived tissue specimens in side-by-side experiments.
Typical research applications include profiling total Raf-1 expression in tumor biopsies, mapping pathway activity in cell lysates from cancer lines treated with kinase inhibitors, and characterizing Raf-1 distribution by IHC in normal and diseased tissue. The polyclonal format recognizes multiple epitopes on the target, which is helpful when probing for total protein levels or when phosphorylation or other post-translational modifications might otherwise compromise recognition of a single linear site.
For handling, keep refrigerated for routine use, and for longer-term storage aliquot and freeze at -20°C to limit repeated freeze-thaw cycles and preserve antibody titer across many experiments. Sodium azide at 0.05% acts as a preservative; downstream applications sensitive to azide, such as certain live-cell or conjugation workflows, may require buffer exchange by dialysis or spin column before use.
