Raised in rabbit against a peptide corresponding to the Q253 region of the Raf-1 kinase, this polyclonal primary antibody from Antibodies.com enables characterization of the canonical RAS–RAF–MEK–ERK signaling pathway. Raf-1, also called C-Raf, is a MAP kinase kinase kinase (MAP3K) that transduces signals from activated RAS GTPases through MEK to ERK, governing cell proliferation, differentiation, and survival programs. The unconjugated reagent is supplied in a vial as 50 µl at 1 mg/ml in phosphate buffered saline containing 0.05% sodium azide at approximately pH 7.2, and is validated for western blot and immunohistochemistry on human, mouse, and rat specimens.

Typical applications include measuring endogenous Raf-1 levels in cell and tissue lysates by western blot and visualizing its distribution in fixed tissue sections by immunohistochemistry. Because the antibody is polyclonal, it recognizes multiple epitopes on the kinase, which can be helpful when probing conformationally distinct or post-translationally modified forms. The reagent is for research use only and supports studies of receptor tyrosine kinase signaling, oncogenic Ras pathway activation in tumor models, and downstream effects on cell fate decisions in normal and disease contexts.
