RAD51D is one of five RAD51 paralogs that operate in the homologous recombination pathway for repairing DNA double-strand breaks, and this unconjugated polyclonal antibody from Antibodies.com detects the human RAD51D protein in Western blot (WB) and immunohistochemistry (IHC) applications. Supplied as 100 µl of rabbit host reagent, the antibody supports immunoblotting of cell or tissue lysates and chromogenic or fluorescent staining of formalin-fixed paraffin-embedded sections.

Functionally, RAD51D acts within the BCDX2 subcomplex together with RAD51B, RAD51C, and XRCC2, where it contributes to the loading and stabilization of RAD51 filaments at sites of DNA damage. Germline mutations in RAD51D have been classified as moderate-penetrance risk alleles for hereditary breast and ovarian cancer, and altered RAD51D expression has been reported in ovarian, breast, and head-and-neck tumor series. The protein is therefore of broad interest in DNA damage response research, Fanconi anemia pathway studies, and synthetic lethality investigations involving BRCA1/2-deficient backgrounds and PARP inhibitors such as olaparib.

Researchers use this reagent to profile RAD51D expression across tumor microarrays, to monitor induction of the protein after treatment with DNA-damaging agents including cisplatin or ionizing radiation, and to compare homologous recombination factor levels between BRCA-proficient and BRCA-deficient cell line panels. The unconjugated format provides flexibility for pairing with standard anti-rabbit secondary antibodies and tyramide-based or HRP/DAB signal amplification systems in IHC, as well as with chemiluminescent or fluorescent detection reagents in WB. Aliquot the reagent upon receipt to limit freeze-thaw cycles and preserve antibody performance across multiple experiments.
