This rabbit polyclonal IgG antibody from Antibodies.com specifically recognizes RAC1 phosphorylated at serine 71 (pS71), a post-translational modification that regulates the small GTPase's membrane association and downstream effector coupling within the Rho family of GTPases. Raised in rabbit and supplied as an unconjugated IgG preparation, the reagent is provided as 100 µl of liquid stock and is validated for western blot detection of endogenous phospho-RAC1 in cell and tissue lysates from human, mouse, and rat samples.

RAC1 operates as a binary molecular switch that cycles between an active GTP-bound state and an inactive GDP-bound state, controlling actin cytoskeleton remodeling, lamellipodia and membrane ruffle formation, cell migration, adhesion turnover, and superoxide production via the NADPH oxidase complex. Phosphorylation at S71, located within the polybasic region immediately upstream of the C-terminal CAAX prenylation motif, modulates electrostatic membrane binding and has been documented downstream of protein kinase A, Akt/PKB, and certain growth factor and integrin cues, as well as during platelet activation. Tracking pS71 therefore provides a direct biochemical readout of RAC1 pathway activation that complements GTP-loading assays.

For western blot, 20–30 µg of total protein separated by SDS-PAGE and probed with an HRP-conjugated secondary yields the expected band at approximately 21 kDa. The antibody has been applied to study RAC1 signaling in fibroblasts, neuronal cultures, and immune cell populations, and cross-reactivity with human, mouse, and rat RAC1 supports comparative work across standard model systems. Store refrigerated at 2–8 °C, protect from light, and aliquot upon receipt to limit freeze-thaw cycles and preserve phospho-epitope binding activity over the working shelf life.
