Paxillin (encoded by the PXN gene) is a 68 kDa focal-adhesion scaffolding phosphoprotein that recruits structural and signaling molecules to integrin-based contact sites. It carries four C-terminal LIM domains responsible for focal-adhesion targeting and an N-terminal region enriched in LD motifs and SH2/SH3-binding sites that undergo tyrosine phosphorylation by FAK and Src-family kinases during cell spreading and migration.
This rabbit polyclonal anti-PXN reagent from Antibodies.com is provided as an unconjugated IgG preparation in a 100 µl volume, intended for Western blot (WB) and immunohistochemistry (IHC) workflows on human samples. The polyclonal format yields recognition of multiple epitopes across the paxillin molecule, which can be useful when probing phosphorylated variants or truncated fragments generated during adhesion signaling studies.
Typical research applications include mapping focal-adhesion dynamics in migrating or spreading cells, characterizing integrin- and growth-factor-dependent signaling cascades, profiling cytoskeletal remodeling in tumor invasion models, and validating CRISPR- or RNAi-mediated knockdown of PXN by Western blot or tissue-section staining. The unconjugated format allows pairing with a secondary antibody matched to the user's preferred detection chemistry, and the IgG isotype permits HRP-, alkaline-phosphatase-, or fluorophore-conjugated secondaries to be selected independently.
Storage follows standard polyclonal-antibody handling: aliquot on first use to avoid repeated freeze-thaw cycles, and protect from light to preserve the unconjugated IgG fraction. For Western blot, RIPA or Laemmli lysates from cultured human cells are commonly loaded in the low-microgram range; for IHC, antigen retrieval conditions are typically required and should be optimized on the target tissue.
