Produced in rabbit as a polyclonal IgG reagent and supplied as an unconjugated 100 µl aliquot, this Antibodies.com primary antibody selectively recognizes PTPN11 (also called SHP-2) only when phosphorylated at tyrosine 580. The Y580 site lies within the C-terminal tail of SHP-2, and phosphorylation at this residue relieves autoinhibition of the phosphatase, an event that couples growth-factor and cytokine receptors to RAS-MAPK signaling during normal proliferation, differentiation, and developmental processes. Gain-of-function mutations in PTPN11 drive Noonan syndrome and LEOPARD syndrome, and the same pathway is implicated in juvenile myelomonocytic leukemia, making phospho-Y580 detection a useful readout of pathway activation in tissue sections. Validated for immunohistochemistry on human, mouse, and rat samples, the antibody enables researchers to map SHP-2 activation in fixed tumor and developmental tissue, supporting research use only investigations into receptor tyrosine kinase signaling, RASopathy mechanisms, and phospho-proteomic profiling of patient-derived specimens.
