Raised in rabbit as an unconjugated polyclonal IgG, this primary antibody recognizes human Prothymosin alpha (PTMA), a 12 kDa intrinsically disordered nuclear polypeptide encoded by the PTMA gene (UniProt P06454). PTMA operates as a chromatin-remodeling cofactor that supports transcription of proliferation-linked genes and protects cells against apoptotic stress, and published immunofluorescence studies consistently place it in the nucleoplasm, with a punctuated distribution that overlaps transcription sites and partially coincides with PML-type nuclear bodies.

The reagent has been validated for IF on cultured human cells and is supplied as a 100 µl liquid vial in a glycerol-containing buffer. It is intended for indirect immunofluorescence: a standard workflow is to fix cells in 4% paraformaldehyde (10-20 minutes at room temperature), permeabilize with 0.1-0.5% Triton X-100 in PBS, block, incubate with the primary antibody overnight at 4 °C, and then detect with a fluorophore-conjugated anti-rabbit secondary such as Alexa Fluor 488, with DAPI used as the nuclear counterstain.

Because the polyclonal reagent recognizes multiple epitopes across PTMA, it can help sustain signal when a single epitope is masked by crosslinking fixation. For best stability, aliquot the vial on first thaw and store at -20 °C, avoiding repeated freeze-thaw cycles.
