This rabbit polyclonal antibody from Antibodies.com specifically recognizes PTK2 (also known as focal adhesion kinase, FAK) only when phosphorylated at tyrosine 397. The phospho Y397 epitope corresponds to the major autophosphorylation site that drives FAK activation downstream of integrin engagement and growth factor receptor signaling.

Validated for Western blotting, the reagent is supplied as 100 µl of unconjugated IgG and is reactive with human, mouse, and rat samples, allowing researchers to track endogenous levels of the activated kinase across standard cell line panels and primary cultures. Typical applications include monitoring adhesion-dependent signaling, probing focal adhesion turnover in migrating cells, and comparing FAK activation states in tumor lysates and tissue extracts, where PTK2 activity is frequently associated with an invasive phenotype and cytoskeletal remodeling.

Because the reagent is phospho-specific, lysate preparation should include phosphatase inhibitors to preserve the Y397 modification prior to SDS-PAGE and transfer. The polyclonal format gives broader recognition of epitopes surrounding the phosphotyrosine than a monoclonal reagent, which is useful when comparing activation across cell lines, stimulation conditions, or pharmacological inhibitors of focal adhesion signaling.
