Raised in rabbit and affinity-purified through an epitope-specific phosphopeptide immunogen that spans the C-terminal regulatory cluster of human PTEN, this unconjugated IgG reagent from Antibodies.com selectively recognises the tumour suppressor only when Ser380, Thr382 and Thr383 are simultaneously phosphorylated. The S380/T382/T383 motif lies within the C-terminal tail (residues 380–385) that casein kinase II (CK2) constitutively modifies, driving PTEN from an open, membrane-associated, lipid-phosphatase-active conformation into a closed, autoinhibited state that is more stable but catalytically dampened by roughly 30% in PIP3 turnover. Detecting that phosphorylated, closed-state pool in cell lysates and tissue sections is therefore a readout of CK2 input onto PTEN rather than of total PTEN abundance. The antibody is supplied as a 50 µl liquid aliquot, resolves the expected ~54 kDa PTEN band, and is validated for western blot, immunohistochemistry and immunofluorescence on human, mouse and rat samples. Store the vial frozen at –20 °C in aliquots to limit freeze-thaw cycles; short-term handling at 4 °C is fine for working dilutions. The reagent is intended for research use only.
