This phospho-specific rabbit polyclonal IgG reagent recognizes PTEN when serine 380 and threonines 382 and 383 are phosphorylated. The clustered C-terminal phosphorylation state is generated primarily by casein kinase 2 (CK2) and is associated with a stabilized, predominantly cytoplasmic PTEN population, reduced membrane recruitment, and lower lipid-phosphatase activity. This selectivity supports research into phosphorylation-dependent PTEN regulation, intracellular localization, PI3K/AKT signaling, and tumor-suppressor function.

Supplied by Antibodies.com as 100 µl of unconjugated antibody, the reagent reacts with human, mouse, and rat samples and supports Western blot, immunohistochemistry, and immunofluorescence workflows. Researchers can compare phospho-PTEN abundance between treated and untreated cell cultures, experimental and control tissues, or relevant disease models. Immunoblotting provides relative phosphorylation measurements, while tissue staining and fluorescence microscopy reveal where the modified protein occurs within cellular compartments. Including a total-PTEN readout and downstream markers such as phospho-AKT can help distinguish changes in PTEN phosphorylation from changes in overall PTEN expression or pathway activation. Because the antibody is unconjugated, an appropriate labeled secondary reagent is required for most detection systems. Phosphorylation-sensitive results should be confirmed with suitable positive and negative controls and empirically validated sample-preparation conditions.
