PTEN (Phosphatase and tensin homolog) is a critical tumor suppressor that negatively regulates the PI3K/AKT signaling pathway by dephosphorylating phosphatidylinositol 3,4,5-trisphosphate (PIP3). Loss of PTEN function is among the most common genetic alterations in human cancers, including endometrial, prostate, glioblastoma, and melanoma, and germline mutations cause Cowden hamartoma syndrome.
