A polyclonal IgG reagent from Antibodies.com provides laboratories with a tool for tracking the Doppel protein (Dpl), the paralogue of cellular prion protein (PrP) encoded by PRND on chromosome 20 approximately 20 kbp downstream of PRNP. Doppel is an N-glycosylated, glycosylphosphatidylinositol (GPI)-anchored membrane glycoprotein that is predominantly expressed in testis but is also ectopically expressed in several tumor types. The unconjugated, affinity-purified antibody is supplied in a 200 µl vial and is validated for Western blot detection of human PRND in cell lysates and tissue homogenates.

On immunoblots, the antibody recognizes Doppel in its characteristic glycosylated form (~30 kDa) and the lower-molecular-weight species (~17-20 kDa) that emerges after glucosidase or PNGase F treatment. The unconjugated format gives laboratories flexibility to conjugate HRP, biotin, or a fluorescent reporter as required by their western blot protocol, which is useful when comparing Doppel and PrP signals side by side or when probing lysates from cells transfected with wild-type or mutant PRND constructs.

Three principal research areas are supported by this reagent. In neuro-oncology, published work on PRT-HU2 and IPDDC-A2 astrocytoma-derived cell lines, and on a 111-sample tissue panel graded by WHO criteria, has shown that PRND/Dpl expression scales with tumor grade and is high in glioblastoma multiforme, anaplastic astrocytoma, gastric adenocarcinoma, and anaplastic meningioma; the antibody is therefore applied in comparative tumor-versus-matched-normal expression profiling and in subcellular fractionation studies distinguishing membrane-associated and cytosolic Doppel pools. In reproductive biology, anti-PRND antibodies have been used to map testis-specific Doppel localization in mouse and pig tissue, including studies on spermatogenesis and Sertoli-cell development. In prion-disease models, the antibody helps resolve Doppel from PrP when probing PRNP-deficient or transgenic cerebella for the ataxia and cerebellar-neuron-loss phenotypes associated with Dpl overexpression.

For best results, store the vial at -20 °C, protected from light, and dispense into small aliquots before freezing to minimize freeze-thaw cycles; centrifuge briefly before opening to recover the full liquid volume. For lysate preparation, centrifuge cleared extracts (e.g. 10,000 x g, 10 min, 4 °C) and load 20-40 µg of total protein per lane on SDS-PAGE for clean Doppel signals on PVDF or nitrocellulose. Intended for research use only.
