Phosphorylation of PRKAA1 (AMPK alpha 1) at serine 496 is a post-translational modification of this central cellular energy sensor, and detecting the modified residue supports research into metabolic regulation, kinase signaling cascades, and downstream growth-control pathways. The Antibodies.com rabbit polyclonal anti-PRKAA1 (phospho S496) reagent was raised against this activating phospho-epitope and selectively recognizes the phosphorylated residue in human samples, giving investigators a tool for AMP/LKB1/CaMKK2 axis studies, type 2 diabetes models, cancer metabolism, and exercise-physiology research.

Supplied as 200 µL of unconjugated IgG, this primary antibody is validated across three immunodetection formats: western blot for resolving the modified AMPK alpha band in lysates from cells treated with AICAR, metformin, phenformin, or glucose withdrawal; immunohistochemistry for mapping spatial distribution in formalin-fixed tissue; and immunofluorescence for subcellular localization in cultured cells. The unconjugated format provides flexibility to pair the reagent with HRP-, biotin-, or fluorophore-conjugated secondary antibodies of the user's choice.

Because the epitope is phosphorylation-dependent, sample preparation matters: lyse cells rapidly on ice in the presence of phosphatase inhibitor cocktails to preserve the S496 mark, and avoid repeated freeze-thaw cycles of the lysate. Aliquot the antibody upon first thaw and store refrigerated in working volumes to maintain titre across multiple experiments.
