This rabbit polyclonal IgG antibody from Antibodies.com selectively detects the PRKAA1 protein only when phosphorylated at serine 496, providing a targeted readout for AMPK alpha-1 activation state in cellular signaling studies. PRKAA1 encodes the catalytic alpha-1 subunit of the heterotrimeric AMP-activated protein kinase complex, a central cellular energy sensor that responds to rising AMP/ADP ratios and to upstream activating kinases such as LKB1 and CaMKK2. The unconjugated format gives researchers flexibility to pair the antibody with custom secondary labels, including fluorescent, HRP, or biotin conjugates chosen for a given detection platform.

Validated applications include Western blot for resolving phosphorylated AMPK alpha-1 from total lysate, immunohistochemistry on formalin-fixed tissue sections for spatial mapping of activation, and immunofluorescence on cultured cells for subcellular localization. The reagent is supplied as 100 µl of polyclonal rabbit serum-derived IgG and is raised against a phosphopeptide corresponding to the S496 site on human PRKAA1. Reactivity is reported for human samples. Aliquot upon first use and store refrigerated to preserve phospho-specific binding activity; repeated freeze-thaw cycles of the working stock should be avoided because phospho-epitope recognition is sensitive to denaturation.

Typical uses include tracking AMPK pathway engagement downstream of metabolic, hypoxic, or pharmacological stimuli such as AICAR, metformin, or A-769662, confirming on-target activity in kinase cascade experiments, and probing cross-talk between AMPK and other serine/threonine kinase pathways. Because the antibody recognizes the phospho-S496 epitope specifically, it complements total-AMPK-alpha antibodies when researchers need to distinguish changes in phosphorylation stoichiometry from changes in total protein abundance.
