PRAS40 (Proline-Rich AKT Substrate of 40 kDa), also called AKT1S1, is a key downstream substrate of AKT kinase and a negative regulator of mTORC1. Phosphorylation at threonine 246 by AKT relieves PRAS40's inhibition of mTOR, coupling this residue to insulin signaling, cell growth, and survival pathways.

The unconjugated rabbit polyclonal antibody from Antibodies.com selectively binds PRAS40 only when phosphorylated at T246, enabling researchers to monitor AKT pathway activation across cell lysates and tissue extracts. Raised in rabbit and supplied at 1 mg/ml, it is formulated in PBS with 0.1% sodium azide and 50% glycerol as a stabilizing cryoprotectant. Validated for Western Blot, the reagent detects the phosphorylated target in human, mouse, and rat samples, supporting studies of metabolic disease, cancer signaling, and neuronal survival.

Each 100 µl vial provides working stock ready for dilution into standard blocking and antibody diluent buffers. The high glycerol content permits frozen storage at -20°C; aliquot upon receipt to limit freeze-thaw cycles and preserve phospho-epitope recognition.
