This rabbit polyclonal antibody targets PPP1R3A (Protein Phosphatase 1 Regulatory Subunit 3A), the skeletal muscle-specific glycogen-associated regulatory subunit of PP1 that directs dephosphorylation of glycogen-bound metabolic enzymes. Raised in rabbit and supplied as an IgG polyclonal at 50 µl per vial, this unconjugated reagent from Antibodies.com is validated for Western Blot detection of the endogenous human and mouse protein.

PPP1R3A localizes to glycogen particles in striated muscle, where it modulates glycogen synthase and glycogen phosphorylase activity; altered expression and coding variants in this gene have been linked to insulin resistance and susceptibility to type 2 diabetes. The antibody recognizes a band of the expected molecular weight in lysates from human and mouse skeletal muscle, supporting studies of glycogen metabolism, metabolic disease models, and the regulation of muscle glycogen handling by exercise or nutrient status.

Suggested western blot workflow: prepare whole-cell or tissue lysates in standard RIPA buffer supplemented with protease and phosphatase inhibitors, resolve 20–40 µg of total protein per lane on SDS-PAGE, transfer to PVDF or nitrocellulose, block with 5% non-fat milk or BSA in TBST, and incubate with the primary antibody followed by an HRP-conjugated anti-rabbit IgG secondary. Signal is developed with chemiluminescent substrate and captured on film or a digital imaging system. Store the vial at -20 °C in working aliquots to limit repeated freeze-thaw cycles and protect from light when not in use.

Research use only. This reagent is intended for laboratory investigation of PPP1R3A in cell and animal models of muscle physiology and metabolic disease, and it is not approved for diagnostic, therapeutic, or clinical use.
