Produced in rabbit and affinity-purified against human DNA polymerase delta, this unconjugated polyclonal IgG primary antibody from Antibodies.com recognizes the 125-kDa catalytic subunit (p125) of Polδ for research use only. The reagent detects endogenous POLD1 across multiple sample types and is supplied as 200 µl of liquid in a vial.

POLD1 provides the essential 5'–3' DNA polymerase activity and the 3'–5' exonuclease proofreading function of the Polδ holoenzyme, which assembles with the accessory subunits POLD2, POLD3, and POLD4 along with PCNA and RFC to drive lagging-strand synthesis, Okazaki-fragment maturation, nucleotide-excision repair, mismatch repair, base-excision repair, and break-induced replication. Loss of POLD1 proofreading activity has been linked to genomic instability and to polymerase-proofreading-associated cancer-predisposition phenotypes, supporting use of this reagent in DNA-replication and DNA-damage-response studies.

Validated for Western blot, immunohistochemistry on paraffin-embedded sections, and immunofluorescence, the antibody reacts with human, mouse, and rat samples. In Western blot the target appears at the predicted molecular weight of approximately 125 kDa. The unconjugated format allows the researcher to select a fluorescent- or enzyme-labeled secondary antibody matched to the chosen detection platform, and the polyclonal IgG formulation broadens epitope coverage of the target. The reagent is shipped refrigerated and should be stored frozen at -20°C in single-use aliquots to limit loss of activity across repeated freeze-thaw cycles; sodium azide is included as a preservative.
