Podocalyxin (PODXL) is a heavily glycosylated type I transmembrane sialomucin of the CD34 family, originally identified as a major component of the glomerular epithelial polyanion in kidney podocytes. It is also expressed on vascular endothelial cells, megakaryocytes, platelets, hematopoietic progenitors, mesothelium, and a subset of neurons, where its strong negative surface charge maintains open filtration slits and supports cell-cell repulsion. Aberrant PODXL expression has been linked to focal segmental glomerulosclerosis (FSGS), diabetic nephropathy, and aggressive epithelial cancers of the breast, colon, stomach, lung, and prostate.

This unconjugated rabbit polyclonal IgG reagent from Antibodies.com detects endogenous PODXL in human, mouse, and rat samples and is validated for Western blot (WB) and immunofluorescence (IF) workflows. On Western blot, the antibody typically recognizes the mature glycosylated form at approximately 160 kDa, well above the 58 kDa predicted molecular weight of the unmodified 558-amino-acid human polypeptide. The polyclonal format recognizes multiple epitopes across the extracellular region, which is useful in tissues where N- and O-linked glycosylation patterns vary between cell types. For IF, the reagent is compatible with paraformaldehyde-fixed cells and cryosections, paired with standard anti-rabbit secondary detection.

Each vial provides 50 µL of liquid antibody, suitable for use as a primary reagent in antibody panels investigating glomerular disease, tumour cell adhesion and metastasis, endothelial activation markers, and early hematopoietic progenitor biology. To preserve activity, store the vial refrigerated at 4°C; for longer-term storage, aliquot and freeze at -20°C, avoiding repeated freeze-thaw cycles. Researchers studying renal filtration, vascular biology, stem-cell markers, and cancer prognosis will find this reagent a useful addition to their workflow.
