Raised in rabbit and affinity-purified, this unconjugated polyclonal antibody from Antibodies.com targets PNPLA2 (patatin-like phospholipase domain-containing protein 2), better known as ATGL (adipose triglyceride lipase), the rate-limiting enzyme that initiates triglyceride hydrolysis within intracellular lipid droplets. Each vial contains 50 µL of IgG preparation, delivered unconjugated for downstream flexibility, and validated for Western blot detection of the endogenous ~55 kDa protein across human, mouse, and rat samples.

PNPLA2 catalyses the first committed step of neutral lipid catabolism, converting stored triglycerides into diacylglycerols for subsequent lipolysis, and its activity is potentiated through direct interaction with the coactivator CGI-58 (also called ABHD5). Loss-of-function variants in the PNPLA2 gene cause Neutral Lipid Storage Disease with Myopathy (NLSDM), an autosomal recessive condition featuring cytoplasmic lipid droplet accumulation in circulating leukocytes (Jordan's anomaly), progressive skeletal muscle weakness, and elevated serum creatine kinase. Investigators working on lipid metabolism, hepatic steatosis, cardiac energetics, brown adipose activation, or muscular dystrophies routinely probe PNPLA2 expression in liver, oxidative myofibers, myocardium, and BAT where the transcript is most abundant.

For Western blot, resolve whole-cell lysates or tissue homogenates on standard SDS-PAGE gels, transfer to nitrocellulose or PVDF, and detect with HRP-conjugated secondary reagents. The polyclonal mixture recognizes multiple epitopes, supporting strong signal under standard chemiluminescent development. Aliquot upon first thaw and store at -20 °C to preserve titer across repeated freeze-thaw events; avoid extended storage in frost-free freezer compartments.
