PNKP (Polynucleotide Kinase 3'-Phosphatase) is a bifunctional DNA repair enzyme central to the base excision repair and single-strand break repair pathways, where its 5'-kinase and 3'-phosphatase activities process damaged DNA termini generated by oxidative lesions, ionizing radiation, and abortive topoisomerase I reactions. This rabbit polyclonal antibody from Antibodies.com was raised against human PNKP and cross-reacts with the mouse and rat orthologs, making it useful for translational DNA damage response studies across standard model systems. Supplied as 100 µl of unconjugated antisera at 1 mg/ml in PBS with 0.1% sodium azide and 50% glycerol, the reagent is validated for western blot detection of endogenous PNKP and for immunohistochemistry localization in fixed tissue sections. Researchers investigating the cellular response to oxidative DNA damage, radiosensitivity, topoisomerase I inhibitor sensitivity, or neurodegeneration-linked PNKP mutations such as MCSZ (microcephaly, seizures, and developmental delay) and Ataxia-Oculomotor Apraxia 4 (AOA4) can use it to track expression levels, subcellular distribution, and recruitment to DNA damage foci. The polyclonal format increases the likelihood of recognizing multiple epitopes on PNKP, which is helpful when working with post-translationally modified or partially degraded protein forms. For short-term use, store aliquots refrigerated; for longer storage, keep frozen and limit freeze-thaw cycles to preserve antibody activity. This primary antibody is intended for research use only.
