Description

A rabbit polyclonal antibody raised against PKC α phosphorylated at Threonine 505, supplied by Antibodies.com as an unconjugated reagent validated for Western blotting (WB) and immunohistochemistry (IHC). The antibody is raised in Rabbit and reacts with Human, Mouse and Rat samples. It is formulated at 1 mg/ml in phosphate buffered saline (PBS) with 0.05% sodium azide at approximately pH 7.2.

The immunogen is a synthetic phosphopeptide derived from human PKC α around the phosphorylation site of Threonine 505, and the antibody specifically detects endogenous levels of PKC α protein only when phosphorylated at Thr505. The reagent has been affinity-purified from rabbit antiserum by affinity chromatography using an epitope-specific immunogen, with a purity greater than 95% as determined by SDS-PAGE. The expected molecular weight of the target is approximately 80 kDa.

Members of the protein kinase C (PKC) family play key regulatory roles in cellular functions including cell growth and differentiation, gene expression, hormone secretion and membrane function. PKCs were originally identified as serine/threonine protein kinases whose activity was dependent on calcium and phospholipids, and are activated by diacylglycerols (DAG) and tumour-promoting phorbol esters. The family can be subdivided into conventional (c) PKC isoforms (α, βI, βII and γ) and novel (n) PKC isoforms (δ, ε, ζ, η and θ); the kinase activities of nPKC δ and ε are independent of Ca²⁺, and the different isoforms show distinct patterns of tissue expression and cofactor dependencies.
