Generated in rabbit hosts, this polyclonal reagent targets Phosphomannomutase 2 (PMM2), the cytosolic enzyme that catalyzes the isomerization of mannose-6-phosphate to mannose-1-phosphate during the early steps of N-linked glycosylation. Loss-of-function variants in PMM2 underlie Congenital Disorder of Glycosylation type Ia (CDG-Ia), making this reagent useful for studies of glycoprotein biosynthesis, dolichol-linked oligosaccharide assembly, and clinical research into glycosylation defects.

Antibodies.com supplies this unconjugated antibody as 100 µl of stock, validated for Western blot (WB) and immunohistochemistry (IHC) applications on human samples. The polyclonal format provides recognition of multiple epitopes on the approximately 28 kDa target protein, supporting detection of endogenous PMM2 in patient-derived fibroblasts, hepatic tissue, and other biopsy material relevant to congenital disorders of glycosylation.

For Western blotting, dilute the antibody in blocking buffer and pair with an HRP-conjugated secondary reagent. For IHC on formalin-fixed paraffin sections, perform citrate buffer antigen retrieval (pH 6.0) and visualize with an HRP-polymer detection system. Refrigerate upon arrival; aliquot to limit freeze-thaw cycles and preserve activity across longitudinal studies of glycosylation pathway proteins.
