PHLDA3 (Pleckstrin Homology Like Domain Family A Member 3) is a p53-regulated tumor suppressor that competes with Akt for binding to membrane phosphatidylinositol (3,4,5)-trisphosphate, thereby restraining PI3K/Akt signaling in the cell. Loss of PHLDA3 expression has been documented in pancreatic, breast, lung, and colorectal cancers, making the protein a useful marker for studying tumor biology, apoptosis regulation, and endoplasmic reticulum stress responses in research settings.

This rabbit polyclonal antibody from Antibodies.com is raised against human PHLDA3 and supplied at 100 µl in an unconjugated format. The reagent is validated for immunohistochemistry on formalin-fixed paraffin-embedded tissue sections and is reactive with human samples, allowing direct detection of endogenous PHLDA3 in archival pathology specimens without the need for a secondary label.

Investigators use this primary antibody to map PHLDA3 expression patterns across tumor tissue microarrays, to correlate staining intensity with p53 status and downstream Akt pathway activity, and to explore the protein's role as an effector of cellular stress signaling in oncology and developmental biology studies. The polyclonal format provides recognition of multiple epitopes on PHLDA3, which can be advantageous when working with partially degraded or fixation-masked antigens.
