Raised in rabbit and affinity-purified to a working concentration of 1 mg/ml in PBS with 0.1% sodium azide and 50% glycerol, this unconjugated primary antibody from Antibodies.com targets Programmed Cell Death 1 (PD-1), the immune checkpoint receptor that engages PD-L1 and PD-L2 to dampen peripheral T-cell activity. Supplied as a 100 µl aliquot, it is validated for Western blot detection of endogenous PD-1 protein in human, mouse, and rat samples.

The reagent binds the cytoplasmic tail of PD-1, a type I transmembrane glycoprotein of the CD28/CTLA-4 family whose ITIM and ITSM motifs recruit SHP-2 phosphatase upon ligand engagement, leading to inhibition of PI3K/Akt and Ras-MAPK signaling in activated lymphocytes. Researchers apply it to profile PD-1 expression on activated T cells, B cells, NK cells, and monocytes within tumor-infiltrating lymphocyte populations, chronic viral-infection models, and autoimmune-disease tissues, supporting studies of checkpoint-blockade immunotherapy, transplant tolerance, and T-cell exhaustion. Store the vial at -20°C in single-use aliquots to limit freeze-thaw cycles, and avoid sodium-azide–containing buffers when planning live-cell staining or downstream conjugation of the antibody.
