Raised in rabbit and supplied by Antibodies.com, this polyclonal reagent targets PCK1 (phosphoenolpyruvate carboxykinase 1, cytosolic), the cytosolic isoform of the rate-limiting gluconeogenic enzyme that converts oxaloacetate to phosphoenolpyruvate in liver, kidney cortex, and adipose tissue. The unconjugated antibody is provided as 50 µl at 1 mg/ml in phosphate buffered saline with 0.05% sodium azide at approximately pH 7.3, and is validated for Western blot detection of the human, mouse, and rat orthologs.

PCK1 expression is frequently read as a marker of hepatic gluconeogenesis, and is of particular interest in studies of fasting, insulin resistance, and diabetes, as well as in tumor metabolic reprogramming where hepatocellular carcinoma and renal cell carcinoma re-engage glucose-producing pathways. The polyclonal format increases the chance of recognizing multiple epitopes on the target, often improving signal when endogenous PCK1 levels are modest in lysates and tissue homogenates.

For Western blot, a starting loading of 20 to 40 µg of total protein per lane, separated by SDS-PAGE and transferred to nitrocellulose or PVDF, is appropriate, with primary antibody dilutions empirically optimized beginning near 1 µg/ml. Brief centrifugation before opening is recommended, and the vial should be stored refrigerated; for use over several weeks, aliquot the solution and freeze at -20°C, avoiding repeated freeze-thaw cycles. The included 0.05% sodium azide acts as a preservative, so the working dilution should not be added to live-cell assays where the preservative would interfere. For research use only.
