The Pax-5 transcription factor (Paired Box 5, also called BSAP) is the master regulator of B-cell identity and commitment. It controls the transition from pro-B to pre-B cell and maintains B-cell lineage fidelity throughout development until the plasma cell stage, when it is actively repressed. Because of this tightly regulated expression window, Pax-5 immunohistochemistry is the standard of care for confirming B-cell lineage in lymphoma diagnosis, particularly for distinguishing classical Hodgkin lymphoma (where Reed-Sternberg cells retain weak to moderate Pax-5 nuclear staining) from T-cell or anaplastic large cell lymphoma.

This unconjugated rabbit polyclonal antibody from Antibodies.com is validated for Western blot and ELISA workflows. The 100 µl aliquot is supplied as a polyclonal reagent raised in rabbit against the Pax-5 target, with demonstrated reactivity against human and mouse samples. The unconjugated format allows researchers to pair it with their preferred secondary antibody, HRP label, or fluorophore for chromogenic, chemiluminescent, or immunofluorescence detection.

Beyond lymphoma diagnosis, researchers use anti-Pax-5 reagents to track normal B-cell maturation in bone marrow and lymphoid tissue, monitor B-cell recovery after bone marrow transplantation, study germinal center B-cell populations, and characterize B-cell developmental defects in knockout mouse models. The paired-box DNA-binding domain is highly conserved across vertebrates, which explains the cross-reactivity with mouse tissue seen here.
