Poly(ADP-ribose) polymerase 3 (PARP3) is a chromatin-associated enzyme central to the DNA damage response, where it is rapidly recruited to double-strand breaks and promotes non-homologous end joining through interaction with DNA-PKcs and the Ku70/Ku80 heterodimer. Produced in rabbit as an unconjugated polyclonal reagent, this antibody from Antibodies.com supports western blot (WB), immunohistochemistry (IHC), and immunofluorescence (IF) workflows with confirmed reactivity against human and mouse PARP3.

PARP3 is a smaller member of the PARP family compared with PARP1, yet it contributes distinct functions in mitotic spindle assembly, telomere maintenance, and class switch recombination in B cells. The antibody recognizes the full-length PARP3 protein (~67 kDa), allowing detection of the target band near this molecular weight in WB lysates of cultured cells or tissue homogenates. Each vial contains 100 µl of antibody; refrigerate on receipt, protect from light, and avoid repeated freeze-thaw cycles to preserve titer.

Typical experimental applications include probing nuclear extracts for PARP3 recruitment to laser-induced DNA damage foci, staining tissue sections to map PARP3 expression in testis and lymphoid organs, and confirming knockdown efficiency in siRNA- or CRISPR-edited cell lines. The unconjugated format allows flexible pairing with HRP-, fluorophore-, or biotin-conjugated secondary antibodies to match a chosen detection platform.
