PARP-1 (Poly(ADP-ribose) Polymerase 1) is a nuclear enzyme central to the DNA damage response and a widely used marker of programmed cell death. The D214 epitope marks one of the principal caspase-3 cleavage sites within PARP-1: when caspase-3 is activated during apoptosis, it cleaves full-length PARP-1 (~116 kDa) at Asp214, generating a large catalytic fragment of approximately 89 kDa and a smaller 24 kDa DNA-binding fragment. Antibodies.com raises this reagent in rabbit as a polyclonal preparation, so the antiserum recognizes multiple epitopes surrounding the D214 region, which can be advantageous for capturing cleaved PARP-1 in denatured lysates.

Validated for Western blot, the antibody cross-reacts with PARP-1 from human, mouse, and rat samples, supporting apoptosis, genotoxic stress, and DNA-damage signaling studies in cell-line models, primary cultures, and rodent tissue. It is supplied unconjugated at 1 mg/ml in PBS with 0.1% sodium azide and 50% glycerol, in a 100 µl vial. The glycerol in the buffer keeps the reagent liquid at -20 °C for long-term storage, while the azide preserves working aliquots kept refrigerated. For Western blot, the antibody produces a clean signal in denatured lysates, allowing visualization of both the intact 116 kDa PARP-1 band and the apoptosis-associated 89 kDa cleavage fragment that serves as a robust readout of caspase-3 activity.
